NEW Systems for Freeze Substitution by Leica Microsystems

15 June 2006: With the Leica EM AFS2, Leica Microsystems presents a freeze substitution (FS) instrument capable of FS and progressive lowering of temperature (PLT) technique as well as allowing low temperature embedding and polymerisation of resins. Intuitive programming and data viewing is via a mouse controlled color screen. A stereo microscope for viewing and positioning of the samples is ALSO available. The LED illumination from within the chamber makes work easy whilst the exhaust system ensures operator safety. LEDs provide the UV light for polymerisation. 
 
Longer processing without interruption 
A 35 l Dewar of liquid nitrogen (LN2) allows protocols over a five day period without the need to refill. Longer processing is no problem as the remote filling allows refilling with LN2 without affecting the temperature of the specimens. Samples can be maintained at temperatures ranging from -140 to +70°C. 
 
Individual user settings 
The ten individual user settings with up to 99 programs each allow numerous parameters to be set and stored. A reagent list is available which can be easily edited. Program data can be transferred from PC to the Leica EM AFS2 via a USB Memory Stick. 
 
New freeze substitution processor 
As an accessory to the Leica EM AFS2 Leica Microsystems offers the new freeze substitution processor Leica EM FSP. It is an automatic reagent handling system that sits on the Leica EM AFS2 and dispenses reagents for both FS and PLT techniques. It automatically dilutes reagents from 100% stock containers thus reducing user set up time and minimising operators contact to toxic media. Through an integrated UV lamp for polymerisation there is only one step for the user – add sample and reagents, press start and leave until end of run. Up to 20 samples can be processed automatically per run in special embedding moulds for high pressure frozen sample carriers and for PLT samples.
