The New Leica EM PACT2 High Pressure Freezer New Standard in Cryofixation by Leica Microsystems
15 June 2006: Leica Microsystems presents the Leica EM PACT2 high pressure freezer allowing high quality freezing up to 200µm into the specimen for subsequent observation in the electron microscope. The Leica EM PACT2 sets the new standard in cryofixation. Mobile and extremely user-friendly Connected to a regular power supply and with its own compressor located on the same trolley it is fully mobile. Interaction with Leica EM PACT2 is via a touch-sensitive color screen where a prompt window makes use very simple. After freezing, pressure and temperature curves can be observed on the screen, then stored and downloaded via a memory stick to PC. Liquid nitrogen (LN2) consumption is very low, requiring around 25 litres per day. Specimen carriers are available for a plethora of various-shaped samples and for a variety of follow-on procedures including freeze substitution, frozen hydrated cryosectioning, cryoplaning and freeze fracture. Transforming high pressure freezing operations with Leica EM RTS With the new Leica EM RTS Rapid Transfer System, Leica Microsystems has introduced an innovative accessory to the Leica EM PACT2 high pressure freezer. The one-step loading allows an ease of sample preparation never before experienced in the field of high pressure freezing. After the user has placed the sample in a carrier, the Leica EM RTS takes over. It locks the sample into a pod and shoots it into the Leica EM PACT2 where it is frozen – all in less than 2.5 seconds. The Leica EM RTS is designed for correlative light and electron microscopy, CLEM, allowing time resolution studies in conjunction with confocal microscopy. A cell can be viewed under the confocal microscope, a system visualised (e.g. protein synthesis using GFP), then taken to the Leica EM RTS where it can be high pressure frozen allowing the same cell to be subsequently viewed in the electron microscope. High pressure freezing at high speed The Leica EM RTS can also be used to enhance the speed of taking a biopsy in conjunction with the new Microbiopsy Transfer system. With a turnaround time of 30 seconds for Leica EM PACT2, samples can be frozen at twice the rate of any other high pressure freezer.
